I would think that if it works on a flash drive, it would be preferable to a hard drive for 2 reasons:
1) Reliability. I've never had a flash drive fail, and I've had many a hard drive bite the dust
2) Portability. (No need to elaborate).
The main difference to me would be load time for an intense Real Track song between the USB flash drive and a USB hard drive. It seems to me the slowest like would be the USB speed itself.
Has anyone done a comparison?

Has anyone tried to use an SD card? Would it be slower or faster than a flash drive? Later, Ray

Hey Matt, it took about 3 hours to load all of the files onto the USB 3.0 thumb drive. Keep in mind write speeds to USB are much slower than read. The program seems to run as fast as it did when I initially installed it on my SSD boot drive.
Also for safety, any files that I create myself are stored in My Documents folder on C drive and auto synched with a cloud service (Sky drive in my case). Jim Hi Jim, Did you run the installer to load the flash drive or just do a copy of the ready to run folder on the hd to the flash drive? Thanks Barry

Handy flash drive tip: Always try plugging in a USB device the wrong way first? If your flash drive (or other USB plug) doesn't have a symbol to indicate which way is up, look for the side with a seam on the metal connector (it only has a line across one side) - that's the side that either faces down or to the left, depending on your port placement.
