The one thing that you will need to do is to make sure that the Guitar Port will work as an input device for the audio out of the computer. Mostly what I would recommend would be a small mixer. Take the outputs of your computer, if you are using a software synth like the Roland VSC DXi and the run them into the mixer. A device like the Guitar Port could be hooked up to your guitar, and then the outputs of that could be fed to the mixer, and then plug in to the mixer.
