You should.
I don't have the 6i6, but with other multi-in/out interfaces I have done this a few times.
Select the second Driver you want for the drum tracks
Audio Preferences-Driver button
Highlight the other driver on the output side (like line out 3/4) and Save
Go to the drum track and assign it to the audio output you just created by using right-click - Port
This tells the track which audio output to use (if it is an audio track).
If it is a MIDI track it'll need to be converted to an audio track, or assigned to a different MIDI output Port ...so it goes to a different synth sound source.
I do not work here, but the benefits are still awesome
