Hi John,
Yes, that is how I run BIAB (I have the Audiophile edition).
There's a file called Readme.rtf in the root directory of the Audiophile's USB drive that explains how to do this. The section of the document that you want to read through is "Options for installing Band In A Box". Options 3 is the one that installs the program and all Realtracks/Realdrums onto your computer's hard drive. This only installs the compressed, wma Realtrack files and not the wav files.
To get the installation happening, just run the setup program in the root directory of the USB drive (it's called Setup.exe) and follow the prompts.
