Sampletank is an instrument, but Amplitube is an effect.

Make sure you have properly registered both at the IK-Multimedia site (you will need your serial numbers to do that using the IK Authorization Manager that you can download. This requires having an IK account set up.

For Sampletank, you need to make sure you match the channel of the MIDI track for which you want to use a Sampletank instrument (if you go to Preferences, Channels, you will see which tracks use which channels). The instrument named on the track won't carry through to Sampletank (that only works for multi-timbral General MIDI Synths, like Coyote WT or VSTSynthFont). In the SampleTank interface, you will need to select the appropriate channel and load an instrument in that slot. You should then hear that instrument play back the MIDI data on the track.

Amplitube is a guitar amp and cabinet simulator. It expects audio output from the track as input to the VST to run it through a virtual guitar amp. It's best to use, for example, one of the DI RealTracks, otherwise it would be like running the sound through multiple amp effects. Select a guitar RealTrack track, load Amplitube, and in the Amplitube interface, select the amp/cab configuration you want.
