Mario’s answer is correct. Theoretically one could make alterations to a RealTrack in Melodyne but that would be painstakingly difficult and tedious. Matt's answer is also correct as I have done that in Melodyne. Also you would need the polyphonic version of Melodyne if you are working with chords. I much prefer to work in MIDI.
