I can hear the background tracks playing, but I am getting nothing from the Thru channel when I try to use my keyboard to play along with. I DO have the Wizard Play Along selected & Wizard to use smart notes selected under the "Play" selection at the top of Biab.

Here is what I was able to find in the F1 "Help" file regarding using the computer QWERTY keyboard the send MIDI to the THRU channel:

MIDI Keyboard Wizard
By turning on the "Use Wizard for THRU part" setting in the Settings for transposing songs when loaded or "Do it Now" dialog, notes played on a Thru channel MIDI keyboard will be played through the Wizard. C, E, G, and Bb will be mapped to chord tones while D, F, A, and B will be passing tones.
Changing Instrument Settings For The Wizard
The Wizard is a play along instrument, so is treated just like the Thru instrument. Changing Instruments/Volume/Reverb etc. for the Wizard is therefore the same as for the Thru instrument. (i.e., Click on the Thru button, then select instrument etc.)
Select the "seeing eye" at the bottom of the audio mixer to open a menu window that lets you choose what channels are visible on the audio mixer.
I've checked the list you provided and they are all selected to use the Thru.
I get a notice when I try and use Asioforall, both A&B and it says they are in use by another program? Yes, I've also tried the MME midi as well to no avail?
I'm stumped because it was working the last time I used Biab, but may have clicked something I shouldn't have? Thanks again for the help.
ASIOforall should not have anything to do with the issue. ASIOforall is a "generic" audio driver and has nothing to do with MIDI.
When you selected the audio mixer "seeing eye" were you able to select the THRU channel for display in the audio mixer?
I noticed in your first screen shot above you have the Cakewalk TTS-1 selected as the Synthsizer\Sound Card and your VSTi|DXi Synth Setting is Cakewalk TTS-1. Try not checking the "Route MIDI THRU to MIDI Driver.
What is the VST MIDI Synth (Port A) and VST MIDI Synth (Port B) displayed in the MIDI Output Driver text box? You do not have a MIDI Output Driver selected.
