It could be a wav file playing back on the single Audio track at the same time, to see, Rightclick on "Audio" trackname and select Mute while song is playing - if the second Melody disappears, then you have created a wav file with the same exact filename as the songfile but with a different suffix. Find that file and remove it from the folder or just choose "Kill Audio" from the Audio menu.

Another thing that can happen is that you ahve somehow inadvertently copied and pasted the Melody to the Soloist track, at another point on the timeline and both are playing back. Check the Soloist and Melody tracks to see if the notation there is like that, if so, you can choose to Kill the Soloist track (or the Melody, whichever one is out of sync) from their corresponding Edit menus. (Again, you can choose each track and RightClick and Mute to find out)

If this continues, first reboot the computer just in case there are lost resources. If still no joy, try the "Return to Factory Settings" in the Options menu inside BiaB.

Finally, there is the off chance that you have a corrupted songfile. Likely the only way to fix that would be to hit New and re-enter the song all over again.
