Covering the simplest things first is just as well. Have you recomposed the tune when starting play?
If you start it using the the "Replay" icon, or by double-clicking on a cell, it will (by design) play again just as it was before you changed the arrangement.
To hear the changes that have been made and make it play in the edited form, start with the "Start" icon or by hitting F4 on the Windows version. There's also the Play menu and maybe other methods.
If you have shut down BiaB in the meantime, this will all take care of itself anyway when you re-start it by any method.
