Amplitube can indeed be invoked on a MIDI sound. I use it, for one example, on the dry Rhodes MIDI voices to add a bit of grit to the Rhodes piano in certain instances, emulating the sound of the Rhodes stage piano when jacked thru, say, a guitar amp.

It is important to understand that the chosen MIDI synth is going to be a DXi or VSTi "instrument" plugin and must therefore be invoked at the top of the list. Then invoke an audio effect such as Amplitube in a slot that is further down the list, making sure that you are now invoking a VST (without the "i" at the end) Audio Effect.
