Well, considering that the Melodist uses MIDI instruments, perhaps you haven't selected a MIDI output device that works, or that device may be turned all the way down in its Control Panel.
Do you see notes in the Notation View on the track where you wanted to place the Melody?
If you see notes that are highlighting but you can't hear them as the song plays, investigate the Options -> MIDI/Audio... settings to see if your MIDI synth is available and that you have selected one.
